ImmunologyOxford Biosystems supplies a wide range of immunoassay kits from various manufacturers for the investigation of the body’s immune response to different disease states and autoimmunity. Autoimmune Diagnostics

Autoimmune diseases are chronic inflammatory processes with indeterminate aetiology, a system of immune responses of an individual against its own healthy cells and tissues. They may be either organ-specific or systemic in nature. Since testing began in the mid 1900’s using Hep-2 cells and tissue sections to detect anti-nuclear antibodies, more than 100 different antibodies have been identified by immunofluorescence and specific immunoassays.

Immunoblot Kits

The DIASPOT range of kits from Diasource employs the immunodot technique which is based on ELISA technology. Nitrocellulose strips enable the detection of up to 12 specific antibodies in a single test and colour-coded reagents are provided ready to use.

The new DIASPOT MULTIQUANT kits are microarrays enabling up to 25 auto-antibodies to be detected and quantified per strip.

The DIASPOT assays can be fully automated in 45 min with the Diasource Neptune Instrument and the Neptune Quantification software will provide quantitative results and assay validation.

DIASPOT kits are ideal for confirming ELISA screening tests, or for running small to medium size workloads.

EliSpot

EliSpot (Enzyme linked immunosorbent spot assay) was originally developed as a method to detect antibody secreting B cells. Later it was adapted to determine T cell reactions to specific antigens, usually reported as the number of activated cells per million. AID GmbH was one of the first companies to design and produce EliSpot readers and ready-to-use kits.

EliSpot System

The iSpot reader enables the analysis of both enzymatic and fluorescent based EliSpot assays. The iSpot reader comprises all the same functionality of the EliSpot Reader Classic, and in addition allows for up to 3 colour FluoroSpot analysis. The patented overlay-technology FluoroAID allows for detailed interpretation of double-expressing T-cells in every well.

AID produces CE-marked 96 well plate assays and 12 x 8 well plate assays. All are provided as complete kits containing pre-coated wells, secondary antibody, conjugate and substrate. Specific antigens are also available.

Immune Response

Oxford Biosystems has a comprehensive range of ELISA kits for the detection and quantification of a wide variety of immune-modulators including cytokines, chemokines, cell adhesion molecules and apoptosis markers.

We also supply ELISA kits for the measurement of specific antibody response to vaccination. These assays are based on the detection of specific IgG and total antibodies (TAb) in serum and plasma and are calibrated against WHO standards where available. The assays are intended for use in the assessment and monitoring of immunisation status and epidemiological studies.

HMGB1 has increasingly attracted the attention of researchers and has become one of the most intriguing molecules within the complete arsenal of proteins of the innate immune system. It is present in the nucleus and cytoplasm of mammalian cells and is involved in gene transcription as a chromatin binder. It can be released passively as a soluble molecule from necrotic cells or may be actively secreted from inflammatory cells, thus it acts like a cytokine with similar pro-inflammatory effects to TNF-alpha and IL-6.

HMGB1 is therefore involved with any diseases coping with dying necrotic cells, examples include, sepsis and septic shock, stroke, cancer and viral infections. There are over 2,000 scientific articles published regarding HGMB1, with over 100 of these having used the IBL HMGB1 ELISA.

IBL International’s HMGB1 ELISA, which has been developed by Shino-Test Corporation, Japan, in cooperation with Prof. Dr. Ikuro Maruyama, Kagoshima University, is the worldwide renowned assay to quantify HMGB1 in any experimental setting.

QUANSYS Q-Plex Array Technology

The Q-Plex™ Array technology is a fully quantitative multiplex ELISA capable of measuring up to 25 distinct analytes per well. Standard kits are available for the measurement of cytokines, chemokines, hormones and other common disease indicating proteins. Multiplex ELISA arrays maximise data generation from each sample and provide a low cost per data point.
