The Value of soluble Interleukin-2 Receptor (sIl-2R) as a Diagnostic and Predictive Biomarker

The interleukin-2 receptor (IL-2R), also known as CD25, is a transmembrane protein, which is up-regulated on activated T-cells, and shed into serum as soluble IL-2R, or sIL-2R. sIL-2R has emerged as an important biomarker that reflects T-cell activation and is often measured in serum to assess immune activity indicating disease status, progression or response to treatment. As serum levels can be increased in various inflammatory conditions, clinical context is required for accurate interpretation.

 

What sIL-2R indicates:

  • T-Cell Activation: It's a key indicator of activated lymphocytes (T-cells), acting as a marker for immune response.
  • Disease Activity: Elevated levels often signal active disease in immune-mediated conditions and cancers.
  • Prognosis: High levels can predict poorer outcomes in certain cancers, like aggressive lymphomas, and severe inflammatory conditions. 

 

Common conditions where sIL-2R may be useful:

  • Cancers: Haematological malignancy and lymphomas
  • Inflammatory Diseases: Sarcoidosis, Rheumatoid Arthritis (RA), Systemic Lupus Erythematosus (SLE), Inflammatory Bowel Disease.
  • Severe Immune Syndromes:  Haemophagocytic Lymphohistiocytosis (HLH). 
  • Multiple Sclerosis (MS): Elevated levels in serum and CSF is indicative of disease activity in MS and strongly associated with relapsing-remitting MS (RRMS) relapses and progressive disease stages.

Measuring sIL-2R provides clinical teams with a sensitive biomarker for monitoring disease activity and treatment response, while research teams may gain valuable insight into T-cell dynamics in inflammatory conditions. 

 

What Is the TECAN-IBL sIL2R ELISA Kit? 

The sInterleukin-2-Receptor ELISA is intended for the quantitative determination of human soluble IL-2 receptor (sIL-2R) in serum and plasma (EDTA, citrate, heparin).

sIL-2R levels can be used as physiological marker for the indication of inflammation in the body, in-vivo immune system activation and response activity. As such the measurement of sIL-2R levels can be applied as an aid in the diagnosis or monitoring of therapy of autoimmune diseases (e.g. sarcoidosis).

The sInterleukin-2-Receptor ELISA is a solid phase enzyme-linked immunosorbent assay (ELISA), based on the on the sandwich principle, and measured on an absorbance reader.

The assay is semi-automated requiring general purpose laboratory instruments and consumables such as absorbance microplate reader/washer, vortexer and pipettes to execute the test. The test kit is intended for manual use and can be adapted to different ELISA processors like TECAN EVOlyzer.

Test results are calculated from a standard curve and compared to laboratory established reference ranges from healthy adults (i.e. normal ranges).
