EliSpot Technology

  • AID GmbH have been making EliSpot assay kits and Reader Systems since 1997. The EliSpot method is not new but only in recent years has it been made usable with the development of EliSpot analyser imaging devices.

     

  • AID GmbH were one of the first companies to offer ready to use EliSpot kits and professionally designed EliSpot readers. All their assays, hardware and software are original company developments.

     

  • AID GmbH is the only fully CE marked company that manufactures EliSpot readers and their systems and software are universally accepted for GMP/GLP situations.

EliSpot Method

EliSpot (Enzyme-Linked ImmunoSpot) is an extremely sensitive immunoassay as it provides detection of secreted cytokines at the single T-cell level. With EliSpot it is possible to detect one specific cell out of 100,000 irrelevant cells, making this method much more sensitive than traditional enzyme linked immunoassay applications.

 

EliSpot was originally developed as a method to detect antibody-secreting B-cells. Later the method was adapted to determine T-cell reaction to a specific antigen. Most researchers use Interferon gamma (IFN-g) production to determine and measure activation of single cells. EliSpot also serves to determine cytotoxic T-cell activity in viral or tumour related studies or to determine Th1-Th2 balance.

 

EliSpot assays are routinely performed in 96-well micotitre plates. Each well contains a membrane coated with specific antibodies i.e. to a particular cytokine. Isolated lymphocytes are incubated with the desired antigen overnight in the well. The antigen activates specific cells to release cytokine, which is captured by the specific antibody. The lymphocytes and antigen are washed away and a secondary antibody labeled with a substrate specific enzyme is added, which binds to the captured cytokine. After washing, the substrate is added and the colour allowed to develop. Analysis of the plate is undertaken using one of the EliSpot Reader Systems.

EliSpot Assays

EliSpots are most commonly used for the measurement of Interferon gamma (also known as Interferon gamma release assays or IGRA), but EliSpot readers can be used to measure many different cytokines and target proteins.

 

EliSpot assays are available as complete, ready to use kits containing a pre-coated 96 well plate, enzyme conjugated secondary antibody and substrate. Kits are available for most human cytokines and customised kits can also be requested.

 

In addition, AID GmbH also provide a selection of ready to use diagnostic kits with specific antigens included (EBV, CMV and Borrelia) and the TransSpot assay, an IFN-g assay for monitoring of transplantation patients and the detection of latent/chronic infections (CMV, EBV, BK virus, Aspergillus plus other antigens to meet patient requirement).

EliSpot Reader Systems

EliSpot Reader classic

EliSpot ClassicCompact (42 cm footprint), efficient and user-friendly, the EliSpot reader interprets any type of EliSpot plate, including all brands of membrane type plates, ELISA-style plates and low volume plates. The EliSpot reader classic simultaneously takes images, auto-centers the well and counts according to the user’s settings. Data acquisition is fully automatic. Counting results can be exported to txt, dbase, Excel, PowerPoint, LIMS, AIDiagnostics or EliStat. Processing time including saving and data export less than 2 minutes per plate. Software and hardware fully compliant with 21CFRpart11, GMP/GLP and CE.

iSpot Reader

Enables analyse of both enzymatic and fluorescent based EliSpot assays. The iSpot reader comprises all the same outstanding functionality of the EliSpot Reader Classic, such as layout-generator, rule-compiler, various export formats and in addition allows for up to 3 colour FluoroSpot analysis. The patented overlay-technology FluoroAID allows for detailed interpretation of double-expressing T-cells in every well. A simple “two-click switch” between fluorescent and enzymatic mode, negates the need for a hardware change. The iSpot Reader is still as fast as previous readers and interprets a 96-well fluorescent plate within 6 to 7 minutes.
